Resumen del proyecto: Chronic lymphocytic leukemia (CLL) is preceded by a condition termed “monoclonal B-cell lymphocytosis” (MBL), in which small clonal B cell subpopulations present in peripheral blood probably expand in response to chronic antigen stimulation, associated to the acquisition of genetic abnormalities, leading to malignant transformation.
SPECIFIC AIMS: 1. To analyze somatic mutations related to clonal hematopoeisis on lymphoid B and myeloid cells from healthy controls, MBLlo, MBLhi and CLL patients; 2.- To study the immune system status in the same 4 groups of individuals; 3.- To identify molecular markers present in B cells and biochemical features realted to antigen afinity against antigens in MBLlo, and MBLhi and CLL patients. 4.- To study the evolution of phenotypic/genetic/molecular features of purified clonal B cells over time, even at the single cell level; and 5.- To integrate all bilogical information with clinical and follow-up data of MBL/CLL subjects.
